Chronic Hyperglycemia Drives Functional Impairment of Lymphocytes in Diabetic INSC94Y Transgenic Pigs

People with diabetes mellitus have an increased risk for infections, however, there is still a critical gap in precise knowledge about altered immune mechanisms this disease. Since diabetic INS C94Y transgenic pigs exhibit elevated blood glucose and stable phenotype soon after birth, they provide favorable model to explore functional alterations of cells early stage vivo . Hence, we investigated peripheral mononuclear (PBMC) these compared non-diabetic wild-type littermates. We found 5-fold decreased proliferative response T tg polyclonal cell mitogen phytohemagglutinin (PHA). Using label-free LC-MS/MS, total 3,487 proteins were quantified, distinct changes protein abundances CD4 + early-stage detectable. Additionally, significant increases mitochondrial oxygen consumption rate (OCR) higher basal glycolytic activity PBMC pigs, indicating metabolic phenotype. Thus, our study provides new insights into molecular dysregulated triggered by permanent hyperglycemia.
