Chronic Hyperglycemia Drives Functional Impairment of Lymphocytes in Diabetic INSC94Y Transgenic Pigs

چکیده

People with diabetes mellitus have an increased risk for infections, however, there is still a critical gap in precise knowledge about altered immune mechanisms this disease. Since diabetic INS C94Y transgenic pigs exhibit elevated blood glucose and stable phenotype soon after birth, they provide favorable model to explore functional alterations of cells early stage vivo . Hence, we investigated peripheral mononuclear (PBMC) these compared non-diabetic wild-type littermates. We found 5-fold decreased proliferative response T tg polyclonal cell mitogen phytohemagglutinin (PHA). Using label-free LC-MS/MS, total 3,487 proteins were quantified, distinct changes protein abundances CD4 + early-stage detectable. Additionally, significant increases mitochondrial oxygen consumption rate (OCR) higher basal glycolytic activity PBMC pigs, indicating metabolic phenotype. Thus, our study provides new insights into molecular dysregulated triggered by permanent hyperglycemia.

Permanent Neonatal Diabetes in INSC94Y Transgenic Pigs

Mutations in the insulin (INS) gene may cause permanent neonatal diabetes mellitus (PNDM). Ins2 mutant mouse models provided important insights into the disease mechanisms of PNDM but have limitations for translational research. To establish a large animal model of PNDM, we generated INS(C94Y) transgenic pigs. A line expressing high levels of INS(C94Y) mRNA (70-86% of wild-type INS transcripts)...

Non-Diabetic Hyperglycemia Exacerbates Disease Severity in Mycobacterium tuberculosis Infected Guinea Pigs

Hyperglycemia, the diagnostic feature of diabetes also occurs in non-diabetics associated with chronic inflammation and systemic insulin resistance. Since the increased risk of active TB in diabetics has been linked to the severity and duration of hyperglycemia, we investigated what effect diet-induced hyperglycemia had on the severity of Mycobacterium tuberculosis (Mtb) infection in non-diabet...

Functional exhaustion of T lymphocytes in chronic toxoplasmosis.

One of the most characteristic features of many intracellular parasite infections is their chronicity indicating that the host immune system is not capable of eradicating the pathogen. Toxoplasma gondii is the most successful parasite worldwide, infecting an extraordinarily broad range of hosts (endothermic animals and humans) and almost all cell types. Recent studies have revealed that in late...

Management of Hyperglycemia in Diabetic Kidney Disease

the growing incidence and prevalence of diabetes has made a significant impact on the development of diabetic kidney disease (DKD) (1). DKD is among the most frequent complications of diabetes; indeed, diabetes accounts for ~50% of all cases of end-stage renal disease (ESRD) in developed countries (2). Patients often have comorbid diabetes and chronic kidney disease (CKD). Although intensive gl...

Strategies of Hyperglycemia Management among Patients with Diabetic Nephropathy, Renal Impairment, Hypertension and Lipid Disorders

The role of smoking in the development and progression of CKD is well established [7,8]. A lot of studies have showed that smoker diabetics have higher prevalence of MA, macroalbuminuria and reduced GFR compared with their nonsmoking counterparts [7,8]. On the other hand, smoking cessation has been showed to be effective at preventing progression of early nephropathy in subjects with T2DM [9]. ...
